Ionic control of sperm motility and trials for the improvement of pufferfish (Takifugu alboplumbeus) sperm extenders

چکیده
Seminal plasma characteristics, such as ionic composition and pH, are important for sperm maturation further motility. In this work, a first batch of experiments evaluated the effect absence several ions (Na+, K+, Mg2+) in artificial seminal plasma, well pH seawater on activation pufferfish (Takifugu alboplumbeus, named before Takifugu niphobles). all experiments, total motility, progressive curvilinear velocity, straight line average path velocity were measured. The Na+, K+ or Mg2+ did not reduce motility comparison with control. contrast, had strong Samples diluted at 6.5 showed high reduction higher pHs (7.5, 8.5 9.5), inhibition was reversible. Where (used medium) concerned, there no significant differences between control (pH 8.2) 6.5, indicating that spermatozoa can swim acidic environments. Finally, nigericin (that make equal intracellular extracellular pH) added, highest found 7.0, which suggested optimal is 7.0. Our second developed to optimize extenders achieve longer preservation under refrigeration, assessing addition concentrations magnesium NaHCO3 other fish species), different pHs. From extender it observed bicarbonate reduced after 7 days incubation, be concluded use 2 mM part composition, combination measured (7.5), seems best option short-medium term storage species.

Comparison of different extenders and storage temperature on the sperm motility characteristics of Kolbroek pig semen
Maintaining a successful pig artificial insemination programme depends on a number of factors, including evaluation of semen characteristics. This study compared the efficacy of different extenders on the sperm motility of Kolbroek semen during short term storage at 4 °C and 25 °C. Semen was collected from Kolbroek boars using the gloved hand technique and transported to the laboratory for eval...
متن کاملP-139: Ram Sperm Motility Following 48 Hours of Refrigeration: Evaluation of Different Extenders for The Preservation of Ram Semen in Liquid State
Background: The application of laparoscopic method for intrauterine insemination is limited in sheep industry by the high cost of equipment and low pregnancy rate of frozen-thawed spermatozoa. Both disadvantages could be decreased if chilled ram semen is transported to the farms and inseminated through cervix. Semen extender provides the requirements for sperm and affects its motility, viabilit...
متن کاملPathophysiology of sperm motility.
Mammalian spermatozoa acquire the ability to swim during their transit from the testis to the oviduct under the control of several external and intracellular factors. These factors play also a pivotal role in regulating acquisition of hyperactivated motility and during the process of chemotaxis. This review summarizes the involvement of such factors in acquisition and maintenance of sperm motil...
